Influence of macromolecular crowding on the charge regulation of intrinsically disordered proteins

چکیده
The coupling between the ionization and conformational properties of two IDPs, histatin-5 ?-amyloid 42, in presence neutral charged crowders is studied by performing semi-grand canonical Monte Carlo simulations.

Quantitative influence of macromolecular crowding on gene regulation kinetics
We introduce macromolecular crowding quantitatively into the model for kinetics of gene regulation in Escherichia coli. We analyse and compute the specific-site searching time for 180 known transcription factors (TFs) regulating 1300 operons. The time is between 160 s (e.g. for SoxS Mw = 12.91 kDa) and 1550 s (e.g. for PepA6 of Mw = 329.28 kDa). Diffusion coefficients for one-dimensional slidin...
متن کاملInfluence of Sequence Changes and Environment on Intrinsically Disordered Proteins
Many large-scale studies on intrinsically disordered proteins are implicitly based on the structural models deposited in the Protein Data Bank. Yet, the static nature of deposited models supplies little insight into variation of protein structure and function under diverse cellular and environmental conditions. While the computational predictability of disordered regions provides practical evid...
متن کاملEffects of Molecular Crowding on the Dynamics of Intrinsically Disordered Proteins
Inside cells, the concentration of macromolecules can reach up to 400 g/L. In such crowded environments, proteins are expected to behave differently than in vitro. It has been shown that the stability and the folding rate of a globular protein can be altered by the excluded volume effect produced by a high density of macromolecules. However, macromolecular crowding effects on intrinsically diso...
متن کاملIntrinsically disordered proteins: regulation and disease.
Intrinsically disordered proteins (IDPs) are enriched in signaling and regulatory functions because disordered segments permit interaction with several proteins and hence the re-use of the same protein in multiple pathways. Understanding IDP regulation is important because altered expression of IDPs is associated with many diseases. Recent studies show that IDPs are tightly regulated and that d...
